Diskeeper 2008 is awesome, perhaps the best defragmenter available. It runs in the background and defrags all my drives so I don't have to, and even has intelligent I/O detection that avoids interference with other applications that access the drive. The file placement feature also seems to do a great job, the games I play most frequently load much quicker now than they used to. Recommended!
